    A technique of timescale analysis performed directly in the time domain has been developed recently. We have applied the technique to studying rapid variabilities of hard X-rays from neutron star and black hole binaries, gamma-ray bursts and terrestrial gamma-ray flashes. The results indicate that the time domain method of spectral analysis is a powerful tool in revealing the underlying physics in high-energy processes in objects.Comment: 7 pages, 4 figures.     Identification of an MCM4 homologue expressed specifically in the sexual stage of Plasmodium falciparum.

    Mini-chromosome maintenance (MCM) proteins play an essential role in DNA replication initiation. We have isolated a novel gene encoding an MCM-like protein from the human malaria parasite Plasmodium falciparum using the vectorette technique. The gene has no introns and comprises an open reading frame encoding 1005 amino acid residues with a predicted Mr of 115 kDa. The encoded protein, termed PfMCM4, contains all conserved sequences in the MCM family and displays the highest homology to the Cdc54 (MCM4) of Saccharomyces cerevisiae. However, PfMCM4 possesses five unique amino acid inserts with sizes ranging from seven to 75 residues. Southern blotting of genomic DNA digests and chromosomal separations showed that the Pfmcm4 gene is present as a single copy per haploid genome and is located on chromosome 13. A 4000-nucleotide transcript of this gene is expressed specifically in the sexual erythrocytic stage, indicating that PfMCM4 may be involved in gametogenesis in which DNA is quickly replicated

    Biodegradation and biotransformation of wastewater organics as precursors of disinfection byproducts in water

    Laboratory experiments were carried out to investigate wastewater organics as the precursors of disinfection byproducts (DBPs) in drinking water supply. The focus was on the change in wastewater DBP precursors during biological degradation under simulated natural conditions. The wastewater and its treated secondary effluent were characterized for DBP formation potential (DBPFP) and DBP speciation profile, including trihalomethanes, haloacetic acids, chloral hydrate, and nitrogen-containing DBPs. Several model organic compounds, including humic acid, tannic acid, glucose, starch, glycine, and bovine serum albumin (BSA), were used to represent the different types of organic pollutants in wastewater discharge. The results show that the DBPFP of wastewater decreased after biodegradation, but the remaining organic matter had a greater DBPFP yield with chlorine. Different model organics displayed different changes in DBPFP during biodegradation. The DBPFP remained largely unchanged for the glycine solution, decreased greatly for the tannic acid and BSA solutions, and increased nearly 3-fold for the glucose and starch solutions after 10d of biodegradation. Meanwhile, the DBPFP yield increased from 3 for glycine to 51μg DBP mg-1 C for its degradation residue, and from 1 for glucose and starch to 87 and 38μg DBP mg-1 C for their organic residues, respectively. Although biodegradation may effectively remove some DBP precursors, biotransformation during the process produces new DBP precursors in the form of soluble microbial products (SMPs).     Factor V Leiden and thrombosis in patients with systemic lupus erythematosus: a meta-analysis.

    The aim of this study was to perform a meta-analysis of the association between the factor V Leiden polymorphism (FVL) and thrombosis among patients with systemic lupus erythematosus (SLE) and/or antiphospholipid antibody (aPL) positivity. Included studies recruited patients based on SLE or aPL-positive status, confirmed subjects' SLE diagnosis as defined by the American College of Rheumatology, and documented thrombotic events. Excluded studies were non-English or considered only arterial thrombosis. Individual patient data, available from 5 studies, together with unpublished data from 1210 European-American SLE patients from the UCSF Lupus Genetics Collection genotyped for FVL, were further analyzed. Seventeen studies (n=2090 subjects) were included in the initial meta-analysis. Unadjusted odds ratios (OR) were calculated to assess association of FVL with thrombosis. The OR for association of thrombosis with FVL was 2.88 (95% confidence interval (CI) 1.98-4.20). In the secondary analysis with our individual patient dataset (n=1447 European-derived individuals), SLE subjects with the FVL polymorphism still had more than two times the odds of thrombosis compared to subjects without this polymorphism, even when adjusting for covariates such as gender, age and aPL status. SLE and/or aPL-positive patients with the FVL variant have more than two times the odds of thrombosis compared to those without this polymorphism

    Fast Locality-Sensitive Hashing Frameworks for Approximate Near Neighbor Search

    The Indyk-Motwani Locality-Sensitive Hashing (LSH) framework (STOC 1998) is a general technique for constructing a data structure to answer approximate near neighbor queries by using a distribution H\mathcal{H} over locality-sensitive hash functions that partition space. For a collection of nn points, after preprocessing, the query time is dominated by O(nρlogn)O(n^{\rho} \log n) evaluations of hash functions from H\mathcal{H} and O(nρ)O(n^{\rho}) hash table lookups and distance computations where ρ(0,1)\rho \in (0,1) is determined by the locality-sensitivity properties of H\mathcal{H}. It follows from a recent result by Dahlgaard et al. (FOCS 2017) that the number of locality-sensitive hash functions can be reduced to O(log2n)O(\log^2 n), leaving the query time to be dominated by O(nρ)O(n^{\rho}) distance computations and O(nρlogn)O(n^{\rho} \log n) additional word-RAM operations. We state this result as a general framework and provide a simpler analysis showing that the number of lookups and distance computations closely match the Indyk-Motwani framework, making it a viable replacement in practice. Using ideas from another locality-sensitive hashing framework by Andoni and Indyk (SODA 2006) we are able to reduce the number of additional word-RAM operations to O(nρ)O(n^\rho).Comment: 15 pages, 3 figure

    Primary structure and sexual stage-specific expression of a LAMMER protein kinase of Plasmodium falciparum.

    We have isolated a LAMMER-like gene from Plasmodium falciparum by vectorette technique. The gene consists of 3316 bp encoding a protein 881 amino acids with a predicted molecular mass of approximately 106.7 kDa. The encoded protein, termed PfLAMMER, is composed of two distinct domains. The N-terminal domain is not related to any previously described protein kinases and has several interesting features including multiple consensus phosphorylation sites for a range of protein kinases, a number of RS/SR dipeptides, a large proportion of charged amino acids, two putative nuclear localisation signals and 14 copies of a tetramer DKYD repeats. The C-terminal domain is characteristic of a kinase in the LAMMER family with the highest homology to the Arabidopsis thaliana AFC3 kinase. Genomic restriction analysis showed that PfLAMMER is encoded by a single copy gene in the parasite genome. A single transcript of approximately 3800 nucleotides is expressed specifically in the sexual stage, indicating that PfLAMMER may be important in regulating the processes of sexual differentiation of the parasite

    A review of physical supply and EROI of fossil fuels in China

    This paper reviews China’s future fossil fuel supply from the perspectives of physical output and net energy output. Comprehensive analyses of physical output of fossil fuels suggest that China’s total oil production will likely reach its peak, at about 230 Mt/year (or 9.6 EJ/year), in 2018; its total gas production will peak at around 350 Bcm/year (or 13.6 EJ/year) in 2040, while coal production will peak at about 4400 Mt/year (or 91.9 EJ/year) around 2020 or so. In terms of the forecast production of these fuels, there are significant differences among current studies. These differences can be mainly explained by different ultimately recoverable resources assumptions, the nature of the models used, and differences in the historical production data. Due to the future constraints on fossil fuels production, a large gap is projected to grow between domestic supply and demand, which will need to be met by increasing imports. Net energy analyses show that both coal and oil and gas production show a steady declining trend of EROI (energy return on investment) due to the depletion of shallow-buried coal resources and conventional oil and gas resources, which is generally consistent with the approaching peaks of physical production of fossil fuels. The peaks of fossil fuels production, coupled with the decline in EROI ratios, are likely to challenge the sustainable development of Chinese society unless new abundant energy resources with high EROI values can be found
